Project Description
The research in this project is focused on structural biology and biophysics of protein interactions in neuronal synapse function and in regulation of synapse loss in Alzheimer’s disease (AD) by the complement system (see e.g Chernyaeva et al. EMBO Reports (2023)). We have long standing interest in complement research (e.g. Kajander et al. PNAS (2011); Bhattacharjee et al. J Biol Chem (2013)), and synaptic adhesion (e.g. Karki et al. Sci Reports (2020), Kim et al. PNAS (2022)). Other projects in the group focus currently e.g. on endoplasmic reticulum stress response regulation, and overall on the HiLIFE protein crystallization core facility operation and related projects.
The successful candidate will work on protein production and molecular interaction studies of complement proteins protective of neuronal synapses in AD, in collaboration with Dr. Karita Haapasalo’s group at the medical faculty. Specifically, protein-protein interactions of complement proteins and ApoE and synaptic proteins will be studied biophysically, biochemically, and by structural biology methods to visualize at the atomic level the protein interactions involved in the complement function at the synapse in disease states.
About you

• Prior experience in protein biochemistry and expression and purification (FPLC) and molecular biology is essential

• High interest in learning/knowledge of structural biology techniques (X-ray crystallography and CryoEM)

We are seeking a doctoral researcher to join the Photosynthetic Microbes group led by Professor Yagut Allahverdiyeva. The positions are part of the PhotoFactory consortium, which has recently received > 2.2 Mil EUR from the ErkkoFoundation. About the Photosynthetic Microbes Research Group: We are studying the regulation of photosynthesis and applying photosynthetic principles to develop sustainable biotechnologies. The research areas encompass various aspects, including the understanding of alternative electron transport routes, utilizing photosynthetic microbes as biocatalysts for production of solar chemicals (including photoH2), biophotovoltaics (BPV) and developing solid-state photosynthetic cell factories, as well as large-scale algae cultivation for phytoremediation and the production of solar chemicals (such as biostimulants and biopesticides). Our group is pioneering cutting-edge technology for photosynthetic production of chemicals and fuels using 3-D printed biocatalysts, also known as solid-state photosynthetic cell factories. For this purpose, we are developing intelligent matrices comprising photosynthetic cells and functional nanomaterials integrated with a network of sensors for monitoring and controlling photosynthetic production. The PhD student will play a pivotal role in establishing infrastructure for AI-assisted cultivation and developing protocols for data collection, processing, and digitalization, enabling data-driven modelling and optimization. The Doctoral researcher will be appointed for a fixed-term 2-year contract (start date: 1 September 2024 or as agreed) to work in a project entitled “HealthFerm- Innovative pulse and cereal-based food fermentations for human health and sustainable diets” more information at https://www.healthferm.eu/ . Fermented foods are widely consumed in Europe and across the globe because of their sensory characteristics, cultural value and – especially in the past few decades – nutritional and perceived health benefits. However, knowledge on the impact of food fermentation on human health and mechanisms behind such impact is limited due to the lack of dedicated and integrated studies. Such knowledge is required to start designing fermented foods with optimal health benefits instead of relying on spontaneous fermentation processes. Such designed foods will preferentially be plant-based (grain-based) as a shift from diets rich in animal-based products to plant-based diets is key in transitioning towards a more sustainable global food system. Fermentation technology provides opportunities to overcome the challenges associated with the incorporation of plant-based raw materials and, hence, supports the required shift. Rational design will require novel microbial resources and rethinking the approach towards food fermentations.

Description Of Position
You, as a mass spectroscopy specialist in the Structural Biology and Analytics unit, will be responsible for the setting up and use of techniques like AS-MS, native MS and HDX-MS to study and understand the mode of action of proteins and modulation of protein actions by small molecules.
This is a permanent position and location of the position is in Espoo.
Description Of Unit
The unit “Structural Biology and Analytics” is a part of the Medicine Design in Innovative Medicines Business Division. Besides the analytical support for all medicinal chemistry activities, the unit focuses on hit finding activities, the understanding of protein modes of action and protein ligand interactions. On site, MS as well as NMR techniques are used to obtain the necessary information. Other techniques like cryoEM or X-ray are used via a strong external network.

Requirements
Practical skills with protein-related MS techniques, especially native MS, HDX-MS or cross-linking techniques, are mandatory to support our activities in the characterisation of protein-ligand interactions and to analyse conformational changes of proteins. The candidate’s special focus will be execution of experiments and data analysis as well as communicating the results to the project team.
The candidate needs to have a good knowledge of protein handling (production beneficial) and must be able to discuss in multidisciplinary project teams the results and the impact of the results on drug design. A basic understanding of biophysical methods is beneficial. A good network with the protein MS community around the world is a big plus.

Etsimme Teknikkoa Surgery/Imaging tiimiin In Vivo (IV) -yksikköömme Kuopion toimipisteeseen. Etsimme ensisijaisesti säteilytyön osaajaa. Tehtävä on määräaikainen.

Säteilytyöhön liittyvät toiminnot o Säteilytyöturvallisuus o Pipetointi ja aktiivisten aineiden käsittely o Punnitseminen o Liuosten valmistus o Radiolaboratorion ja HPLC tilojen toimintaan ja laitteisiin liittyvät ylläpitotoimet o Aktiivisen jätteen käsittely, varastointi ja hävitys o Erilaisten laitteiden käyttö (Gammacounter, annoskalibraattori, PET/CT, SPECT/CT, taustasäteilymittari/dosimetrit)
Edellytykset
