Subject field and the description of the position
Applications are invited for a full-time PhD student position for a maximum of four years, starting on January 1st 2020, in a Biocenter Oulu research project headed by Professor
Petri Kursula in the Biomedical Structural Biology research unit, Faculty of Biochemistry and Molecular Medicine.
Our
main focus is the structural biology of the myelin sheath. Most myelin
proteins are either integral or peripheral membrane proteins, often
involved in either autoimmune or inherited
demyelinating disease. We also have a keen interest in other structural
biology problems, including protein-membrane interactions at the
molecular level, high-resolution crystallography, and the structural
basis of various neurological diseases. More details

The
position is supervised by Prof. Petri Kursula and Dr. Salla Ruskamo and
is focused on the 3D molecular and atomic structure of the myelin
sheath, as well as the molecular mechanisms
of mutations causing human hereditary neuropathies.
Required Qualifications and assessment
To
successfully occupy the PhD student position, the candidate must
possess an MSc degree in biochemistry, biophysics, protein science, or
another relevant field. Practical experience
in protein research will be highly valued, and a keen interest in
biophysics and structural biology are required. The MSc degree must have
been completed with good grades by December 2019, preferably by the
call deadline. Ability to pursue independent research
as a member of a team and excellent oral and written communication
skills in English are expected.
The
position is intended for an Early Stage Researcher (ESR) who, according
to the ”European Charter for Researchers and the Code of Conduct for
the Recruitment of Researchers”, is defined
as a researcher in the first four years (full-time equivalent) of
his/her research activity, including the period of research training.
Applicants will be either starting their doctoral training, or will have
completed no more than two full years of training
(adjusted for possible previous part-time status) at the time the
position is filled. The selected candidate will carry out his/her
doctoral studies in the HBS-DP of the UniOGS and must meet the minimal
requirements to receive doctoral study rights as defined
in http://www.oulu.fi/uniogs/requirements_for_admission.
Salary
The
salary of the selected doctoral student will be initially set on level
2, and later on levels 2 – 4, of the national salary scale for the
teaching and research staff of Finnish
universities. In addition, a supplementary remuneration is given for
personal achievement and performance (max. 50 %). In practice, the
salary is roughly 2200 – 2700 € per month for an early stage researcher,
depending on qualifications and progress of the
doctoral thesis.
Since
this call is for a starting PhD student, the position will be filled
for a four-year period, beginning on 1 January 2020. A six-month trial
period will be applied in the position.

Subject field and the description of the position
The FET open project MindGAP is aiming to characterize CNS originating Extracellular Vesicles from the blood of cancer patients undergoing therapy. The project involves EV isolation from biofluids and primary human cell culture, Characterization of EV surface molecule, RNA analysis (ddPCR and Next Generation Sequencing) and collaboration with other members of the consortium, in Portugal and Oulu. We are looking for a candidate with strong molecular and cellular biology background and interest for Extracellular Vesicles. Experience with liquid biopsy, Extracellular Vesicles RNA/ DNA, surface protein isolation, bioinformatics/ neurobiology is an advantage.
The duration of the position is from September 1st 2019, or as soon as possible thereafter, ending in March 31st 2023. Required Qualifications and assessment The successful applicant must hold an MSc degree in life sciences (Biology/ Biochemistry). Candidates whose master thesis will be submitted by the application deadline will also be eligible. Good oral and written English skills, good communication and teamwork skills are required.
A six-month trial period will be effective in the beginning of a contract.

The area
Your will be working within the CVMD iMed Bioscience department, which is a multi-discipline
department
working with metabolic, cardiovascular and kidney diseases. The drug
hunting activities also spans into many different modalities including
small molecules, peptide and nucleotides.
This provides scientists with a unique opportunity for innovation in drug discovery.
You will be part of the metabolism in vitro group in Bioscience and focusing on questions within drug
discovery project in diabetes and NASH areas.
The Cell Biologist Metabolism supports drug discovery
projects with scientific input in the in vitro assay strategy and delivers key in vitro and ex vivo
decision-making data.
Major responsibilities:
• Independently apply existing in vitro and ex vivo assays for projects during different phase of
projects.
• Independently design and execute in vitro or ex vivo experiments
• Statistical analysis and interpretation of generated data
• Presentation of data at the team and project team level
• Document the work according to AstraZeneca standards, and prepare the preclinical
pharmacology part of regulatory documents as appropriate.
• Work collaboratively with in vivo, DMPK, etc scientists as required to ensure flow-scheme
connectivity and translatability between in vivo and in vitro models
• Work collaboratively within Bioscience and iMed to deliver on project key questions.
• Contribute proactively to a vibrant and innovative working environment where improvements
and new ideas are adopted and freely shared.
• Ensure compliance with corporate code of conduct guidance and other safety standards.
Minimum requirements
• Broad expertise in cell biology & biochemistry
• Experienced in establishing/performing cell-based assays.
• Cell culture, ELISA, western blot, and qPCR experience is a must
• Experience with primary cell culture, measurements of cellular respiration, and knowledge of
adipose and liver cell biology is an additional advantage.

spanning the UK, US and Sweden. You’ll work with some of the most
knowledgeable technological experts in the industry, all collaborating
on high-profile drug discovery projects.
Extracellular
vesicles represent an exciting opportunity for delivery of complex
RNA-based drugs, and since 2015 we have been working with a team of
postdocs on developing new technologies for drug loading, targeting and
functional delivery using extracellular vesicles. We now have openings
for three new postdocs working on 1) delivery of mRNA, 2) targeting to
heart, and 3) developing innovative in vitro and in vivo
models of heart disease to explore the action and underlying biological
mechanisms of engineered extracellular vesicles in a therapeutic
setting. The internal postdoc work at AstraZeneca is supported through
collaborations with leading academic experts.
Myocardial
infarction (MI) results in severe damage to the heart muscle tissue with
scar formation and irreversible loss of cardiac function leading to the
development of heart failure. Encouraging results have been achieved
with cell therapy to regenerate the heart tissue and improve function,
but there is accumulating evidence that these beneficial effects are
mainly mediated by extracellular vesicles. We propose to study the
action of engineered extracellular vesicles from human IPS-CPCs (cardiac
progenitor cells) and from adult stem cells in vitro (angiogenesis, cardiomyocyte proliferation, apoptosis) and in rodent models of MI in vivo. We will engineer extracellular vesicles
to carry additional (known and novel) stimulatory factors to augment
their biological activity (e.g. VEGF-A, mRNA coding for genes driving
proliferation, etc.). We have developed novel cell-based and in vitro
technologies for EV loading with proteins and RNA, which will be
exploited in this project (focus for postdoc-1). We will explore targeting of extracellular vesicles to heart tissue (focus for postdoc-2) with known and novel ligands and examine improved drug exposure to heart tissue. We will multiplex in vivo
experiments with extracellular vesicles by bar-coding technology for
bio-distribution studies. In addition, we have a separate recruitment
ongoing in CVRM for a 3rd AZ postdoc working on the development of novel
in vitro and in vivo regenerative models of heart
disease. The three postdocs will work closely together on the shared
goal of developing novel opportunities around the use of engineered
extracellular vesicles for therapeutic treatment of heart disease. As a
postdoc you will also get the opportunity to try novel techniques and
ideas. We will also support development of leadership skills and deeper
understanding of drug development process.
Education and Experience required:
Essential:
- A PhD in biological sciences with a strong cell biology or biochemical focus.
- A strong scientific track record, as evidenced by published papers in high quality peer-reviewed journals.
- Excellent
organizational, communication and interpersonal skills, combined with
the ability to work collaboratively across boundaries and problem solve
in the absence of complete information.
- Entrepreneurial ways of working
Desirable:
- Experience of cell imaging, confocal microscopy, FACS and RT-PCR
- Experience with in vivo experiments and bio-distribution
- Experience with EV drug loading (postdoc-1) and/or drug targeting (postdoc-2)
Skills and Capabilities required:
- Working experience of isolation, characterization and manipulation of extracellular vesicles.
- Excellent
expertise with mammalian cell culture including iPSC and adult stem
cells, general molecular biology techniques, RNA biology, and protein
engineering.
- Experience working with proteomics and transcriptomics data
- Strong oral and written communication skills
