Laboratory of Stem Cells and Cancer‐ Université Libre de Bruxelles
http://blanpainlab.ulb.ac.be/
PhD student position
4 years grant (2 years renewable), starting October 2019
Our laboratory is located on the medical campus (Erasme) of Université Libre de Bruxelles (Belgium) and is headed by Prof. Cédric Blanpain. This project will be performed under supervision of Dr. Alexandra Van Keymeulen and in close collaboration with Dr.Christel Pequeux at Université de Liege.
Research Project
Approximatively 75% of breast cancers are estrogen dependent and estrogen receptor‐positive (ER+). Despite the large efficacy of anti‐estrogen treatments, relapse occurs in 30% of cases. In addition, estrogens remain the most powerful treatment to circumvent menopause symptoms. However, their use as menopause hormone treatment (MHT) remains controversial since MHT is associated to an increased risk of breast cancer. A better understanding of the ER signaling upon estrogen or anti‐estrogen treatment in mammary gland and during breast tumorigenesis is thus crucial to develop new tools to target these cancers and to develop a new generation of MHT.
In this context, we aim at answering the following questions:
Aim 1: What is the impact of estrogen signaling on the transcriptional and chromatin landscapes of
the ER+ and ER‐ epithelial cells during tumorigenesis?
Aim 2: Which mechanisms are promoting breast cancer upon MHT treatment during menopause?
Aim 3: How come estetrol treatment during menopause does not promote breast cancer, while it prevents side effects of menopause?
These experiments will constitute a novel and comprehensive definition of the transcriptional and chromatin landscapes of purified individual cell lineages during tumorigenesis and upon MHT. Our results will constitute the basis to highlight new potential targets to treat luminal‐like breast cancer and will also provide precious information to assess how estetrol could safely replace estradiol use as MHT, without increasing breast cancer risk. Finally, they will open new avenues to prevent hormone‐dependent tumorigenesis.
Methods: in vivo models of mammary gland and breast cancer (MMTV‐PyMT, PDX, cell line xenografts), FACS cell sorting, RNAseq, ATACseq, Chipseq, qPCR, 3D cultures.
Skills: Master degree in Biomedical science, molecular biology, biochemistry or equivalent, FELASA certification. High motivation, enthusiasm and scientific curiosity are mandatory. Ability to learn fast,
to work independently, meticulous, good communication would be appreciated.
Contact: To apply, send a motivation letter, a C.V. and two references with their email address to

The University of Copenhagen is seeking a highly motivated and
talented postdoctoral fellow to commence 1 October 2019 or as soon as
possible thereafter at the Novo Nordisk Foundation Center for Basic
Metabolic Research, Faculty of Health and Medical Sciences.
The postdoctoral fellowship is part of the Novo Nordisk Foundation
Center for Basic Metabolic Research International Postdoctoral Program www.cbmr.ku.dk/postdocprogram.
The postdoctoral fellow will receive a competitive package including
salary and running costs for three years, and be part of an
international scientific collaboration at the Maersk Tower www.maersktower.ku.dk.
Job description
The fellowship is aimed at early career researchers with a basic
science background or clinicians who aspire to a career in academic
medicine. We are particularly interested in candidates who are familiar
with integrative research approaches within the broad area of basic
metabolic research with an application towards human pathophysiology.
The Center’s vision is to strengthen interdisciplinary research that
transforms the basic understanding of mechanisms involved in metabolic
health and disease, and to accelerate this knowledge toward new
prevention and treatment strategies. Information about the Center can be
found at www.cbmr.ku.dk.
The postdoc position is in the Genomics Discovery and Epidemiology
Group headed by Associate Professor Tuomas Kilpeläinen. The research
group investigates how genetic variation contributes to obesity and
related metabolic disorders and interacts with the environment,
modifying the effect of physical activity, diet, and other lifestyle
factors on weight gain and metabolic health. You can read more about the
Kilpeläinen Group at www.cbmr.ku.dk/research/human-genomics-and-metagenomics-in-metabolism/kilpelainen-group.
Project description
Insulin resistance, dyslipidaemia, and increased risk of type 2
diabetes are closely linked to obesity, but there is wide individual
variability in the metabolic tolerance for carrying excess body fat.
Among patients who are morbidly obese, up to 10% do not show any sign of
metabolic dysfunction at all. In contrast, lipodystrophic patients who
are unable to form subcutaneous adipose tissue, typically suffer from
severe insulin resistance and other metabolic abnormalities. Overfeeding
experiments have shown that given similar relative weight gain,
individuals may be either protected from or predisposed to metabolic
dysfunction. Furthermore, results from weight loss interventions for
type 2 diabetes have shown that non-overweight individuals respond to
substantial weight loss just as well as the obese in terms of conversion
to normal glucose tolerance.
The aim of the present project is to examine the role of genetic
variation in the individual relationship between adiposity and metabolic
dysfunction in human populations. We expect that the postdoc will
develop a project that, through the understanding of the role of genetic
diversity, will elucidate the causal mechanisms that couple obesity to
metabolic dysfunction and open avenues for identifying individuals with
the highest risk of developing insulin resistance and type 2 diabetes if
they gain weight.

Transcriptional regulatory mechanisms of integrins, as well as crosstalk between integrin signaling, androgen receptor pathway and other relevant prostate cancer associated pathways will be investigated. Genomic analysis will focus on systems identification of causal genetic variants, causative genes and relevant pathways as well as their functional and biological effects on prostate cancer susceptibility, initiation and progression.
The project utilizes 3D cell culture models where, in addition to established prostate cancer cell lines, we develop novel study systems based on prostate cancer patient-derived prostate organoids. The candidates will also employ advanced microscopy techniques, virus-based genomic engineering, CRISPR-Cas9-mediated genome editing and state-of-art cancer genetics and functional genomics approaches. Two PhD-student positions (application and more information in the link):
https://rekry.saima.fi/certiahome/open_job_view.html?did=5600&jc=1&id=00007583&lang=en
Applications are invited for two full-time doctoral student positions for a maximum of four years, starting at the earliest on 01.01.2020 in a Biocenter Oulu research project headed by Professors Gonghong Wei and Aki Manninen in the cancer genomics research unit, Faculty of Biochemistry and Molecular Medicine.
Our work aims to discover the roles of novel transcription factors and epigenetic regulators in cancer predisposition and progression, and to explore how these proteins misregulate genome-wide gene expression in particular during prostate cancer development. We also focus on the identification of causal genetic variants, genes and relevant pathways as well as their functional and biological effects on prostate cancer susceptibility, initiation and progression.
The positions will be supervised by Professors Gonghong Wei and Aki Manninen and are focused on mapping genome-wide binding sites (cistrome) for novel transcriptional regulators in prostate cancer cells and tissues. We will further investigate the cistrome-enriched prostate cancer risk-associated variants and thus integrate the eQTL, epigenome and three-dimensional (3D) genome data to unravel the biological roles of causal variants and genes. Functional validation of selected genetic variants, target genes and other relevant risk factors will be performed using genetically engineered prostate cancer models. The projects utilize 3D cell culture models, potential patient-derived prostate organoids/xenografts, mouse models, advanced microscopy techniques, viral gene transfer-based genomic engineering, and CRISPR-Cas9-mediated genome editing as well as other molecular cell biology methods and functional genetics/genomics approaches.

PhD fellowship is available at the Faculty of Biosciences and
Aquaculture, Nord University (Bodø, Norway) on the project: Biogenesis
and functions of small regulatory RNAs in embryonic development of
zebrafish.
The position is for 3 years and will be available from August 1st. 2019.
The PhD is in molecular developmental biology, and zebrafish is the
primary model. Regulation of early embryonic development is a
multifaceted process involving many classes of regulatory elements.
Ambiguity of developmental origin of these elements is a major
difficulty in studying their functional roles This study combines
functional genomics and high-throughput sequencing approaches in order
to identify biogenesis, expression features, and roles of chosen
elements. The focus is on two classes of small regulatory RNAs: miRNA
and piRNA, which are both present in the early embryo yet their roles
are poorly determined.
