Suomen kesä on alkanut erittäin lämpimästi ja kesän tuloon liittyy tietysti myös vuoden ensimmäinen Solubiologi. Tämän vuotisen kevään numeron aloittaa Marjaana Parikainen, joka avartaa meille kollageeni I:stä sitovan α2β1-integriinin roolia eturauhassyövässä, ja tämän vaikutusta geeniekspressioon ja invaasioon. Seuraavaksi Teemu Syrjälä kertoo meille brominoituneista palonestoaineista ja niiden vaikutuksista syöttösoluihin. Tämänkertaisen tietopaketin päättää Moona Huttusen ulkomaankirjeenvaihtotervehdys, jossa käydään läpi hänen jo kolme vuotta kestäneitä seikkailuitaan Lontoossa University Collegessa, Englannissa.

Tänä kesänä saimme mahdollisuuden lähteä laboratorioryhmän kanssa joukolla Suomea pakoon vierailulle TuE:hen (Technische Universiteit Eindhoven) Eindhoveniin, Hollantiin, jossa tapasimme ryhmänjohtajamme Cecilia Sahlgrenin toisen ryhmän tiimiläisiä. Ahkerana professorina Cecilia pyörittää siis kahta kokonaisuutta, ja olihan se mielenkiintoista päästä tutustumaan teknillisen yliopiston työtapoihin ja tekniikkoihin. Tämä alunperin 1956 perustettu yliopisto kouluttaa pääasiassa insinöörejä teknologian alalla, mutta solu- ja kudostekniikan ja mikrofabrikoinnin laboratoriot yhdistävät solubiologiaa ja biotekniikkaa. Yhteistyö insinöörien ja solubiologien välillä on tärkeässä asemassa TuE:ssa, kun kehitetään esimerkiksi keinotekoisia sydänläppiä tai mikrosiruja, joilla halutaan simuloida erilaisia olosuhteita, joihin solut voivat joutua. Lopullisena päämääränä on kehittää niin sanottuja organ-on-a-chip –menetelmiä, joilla voitaisiin simuloida kokonaisia elimiä pienoismallissa.

Tämä jäänee omalta osaltani päätoimittajuuteni viimeiseksi Solubiologi-lehdeksi, koska syksyllä puikkoihin tarttuu J.J. Hyvää kesän jatkoa kaikille toivottaa SEb ja koko Solubiologin väki.

1. Artikkeli. Kollageenireseptori α2-integriini säätelee eturauhassyöpäsolujen geeniekspressiota ja invaasiota, Marjaana Parikainen (FM)

Erikoistyössä tutkittiin α2-integriinin vaikutusta eturauhassyöpäsolujen geeniekspressioon sekä solujen migraatioon ja invaasioon neljällä α2-integriiniä erilaisesti ilmentävällä DU145-solutyypillä. α2-integriinin havaittiin säätelevän tiettyjen geenien ilmentymistä sekä RNA- että proteiinitasolla mutta se ei vaikuttanut säätelemiensä proteiinien lokalisaatioon solussa. Lisäksi havaittiin, että α2β1-kollageeni I -vuorovaikutus lisäsi solutyyppien välisiä RNA-tason ilmentymiseroja suurimmalla osalla tutkituista 12 geenistä, minkä perusteella α2-integriinin aktivoituminen ja signalointi ligandivuorovaikutuksessa säätelee näiden geenien ilmentymistä. α2-integriiniä ilmentävät solut invasoivat enemmän Matrigeeliin ja kollageeni I:een kuin α2-integriiniä ilmentämättömät solut. Tulosten perusteella α2β1-integriiniin kohdentuvaa syöpähoitoa voitaisiin mahdollisesti hyödyntää eturauhassyövän luumetastaasien estämiseen.

 

2. Artikkeli. The effects of some common brominated flame retardants (bfrs) on cell degranulation and cell viability in a mast cell line, Teemu Syrjälä (Biomedical Laboratory Scientist, Bachelor of Health Care; EQF6)

The aim of this study was to investigate the effects of two commonly used brominated flame retardants, tetrabromobisphenol A (TBBPA) and hexabromocyclododecane (HBCD), on degranulation and viability of a mast cell line RBL-2H3. Assays measuring and monitoring cell viability and the release of inflammatory proteins via degranulation were developed. The results showed that both TBBPA and HBCD cause substantial cell death over a long-term exposure in vitro. Both compounds caused some degranulation of the cells, although the results from HBCD turned out to be quite modest compared to those gotten from TBBPA. TBBPA greatly induced cell death by apoptosis while only a very small degree of necrotic cell death was observed. While the degree of necrosis caused by HBCD remains unknown, it was shown to also induce apoptotic cell death at least to some extent. The study concludes that TBBPA, based on the results, does seem to be a mast cell sensitizing agent. HBCD has great potential to have similar effects, although this requires further research.

The position is available from 1st September 2018 for one year with the possibility of extension. You will be the part of the multifaceted projects aimed at understanding the role of extracellular vesicles in osteosarcoma development and progression; developing 3D models for osteosarcoma; bone tissue engineering/applications of biomaterials and stem cells. In addition to strong written and oral communication skills, experience with techniques in genetics, cell biology, cell imaging, RNA sequencing, biochemistry and bioengineering are considered favorable.

The discovery of a fatal neurodegenerative disease in childhood with fibrosis, and cerebral angiomatosis (FINCA disease) has highlighted the importance of NHLRC2 in the central nervous system (Uusimaa et al. 2018) (doi: 10.1007/s00401-018-1817-z). We are now looking for a postdoctoral scientist to focus on characterizing the role of NHLRC2 during neurogenesis in a two-year project funded by the Academy of Finland.

THE IDEAL CANDIDATE
We expect you to have a medical or scientific background with established relations to the medical community, and preferably a Ph.D. or other strong background in the relevant therapeutic area. You have experience from the industry in a relevant role and knowledge of the clinical development process. You are capable of comprehending large amounts of scientific content which you can communicate in a clear and concise way and have excellent presentation skills. As a person you are a team player, self-driven, independent, resourceful and trustworthy. You have strong statistical skills with ability to extract insights from data. We expect you to enjoy creating new contacts and have good social skills. You are also excellent at communicating, both verbally and in writing. You are fluent in Finnish as well as English orally and written.

Hallituksen kokous 8.6. Helsingissä

Suomen Solubiologit ry:n hallitus kokoontui perjantaina 8.6. ravintola Zetorissa, Helsingissä. Kokouksessa suunniteltiin syksyn seminaaria, jonka teemoiksi on kaavailtu rohkeutta ja innovatiivisuutta. Yksi täydellisesti teemoihin sopiva puhuja on jo varmistunut, ja tänään saatiin mietittyä muitakin rohkeita solu- ja molekyylibiologian alan toimijoita. Lisäksi suunniteltiin tapahtumaa ensi vuodelle, joka on Suomen Solubiologit ry:n 45v-juhlavuosi! Stay tuned!

Indiana University School of Medicine, Department of Anesthesia, has a federally-funded postdoctoral position open in a cutting-edge Shock, Trauma and Neuroscience Research Laboratory that focuses on immunologic response to traumatic brain injury (TBI), trauma, and hemorrhagic shock.
 
The candidate will lead studies in the following areas:
1) persistent pain due to mild traumatic brain injury in adults: alterations of brain function, central sensitization, and inflammatory processes;
2) monocyte/macrophage transcriptional response to TBI in rodent models of mild TBI and polytrauma;
3) mechanisms of immune modulating therapy on the anatomic, physiologic, and transcriptional response to TBI, polytrauma, and hemorrhagic shock;
4) characterization of a model of mild TBI (concussion) for the long-term study of microglial and macrophage gene expression and response to immune modulating therapy; and
5) precision typing to determine sensitivity and tolerance of TBI and hemorrhagic shock.

Specific Responsibilities
·         Process and analyze clinical blood samples derived from mild TBI patients, and assist in quantitative sensory testing of patient volunteers.
·         Induce experimental traumatic brain injury alone or combined with polytrauma in rodents; harvest and process brain, muscle, bone, and viscera; perform and analyze advanced flow cytometry/cell sorting; administer, and collect and analyze RNA sequencing data.
·         Supervise students and other techs working in the lab.
·         Help write and contribute to publications resulting from research done in the lab.
·         Perform other duties as assigned.
 
Candidates must be highly self-motivated and self-sufficient with PhD or MD. A strong background in immunology and flow cytometry is a requirement and some background in neuroscience and pain would be preferred. Experience with rodent models is ideal. Experience with RNA sequencing is desired.

ASSISTANT PROFESSOR / ASSOCIATE PROFESSOR / PROFESSOR IN NEUROBIOLOGY

The successful applicant may be appointed to a permanent full professorship or a fixed-term associate / assistant professorship (tenure track) depending on their qualifications and career stage.
The professorship is based in the Molecular and Integrative Biosciences Research Programme, which is one of the three Research Programmes in the Faculty. Neurobiology, including electrophysiology and molecular and developmental biology of the brain, is an area of strength of the Programme. The other areas of strength of the Programme are cell and developmental biology, genetics, virology and structural biology (for further information see https://www.helsinki.fi/en/faculty-of-biological-and-environmental-scien…). 

The research and teaching activities at the Viikki Campus are supported by state-of-the-art infrastructures including core facilities for e.g. DNA sequencing and genomics, proteomics, metabolomics, lipidomics, and light- and electron microscopy including two-photon confocal and cryo-electron microscopes. Within the Research Programme, neurobiology research is supported by an in-house animal facility, and electrophysiological instrumentation is abundant in the laboratories. There are numerous research groups in the Viikki Campus that focus on e.g. molecular and developmental neurobiology, mechanisms of cell and network level activity, as well as on neurodevelopmental and neurodegenerative diseases. The Viikki Campus is one of the three centres of neuroscience research in the Helsinki area, the other two being located in the Meilahti Campus, and at the Aalto University. 
The professor is expected to contribute to the advancement of neurobiology in the Molecular and Integrative Biosciences Research Programme. Demonstrated excellence and scientific leadership in the field of neurobiology is a requirement.

The main tasks of the assistant professor / associate professor / professor in neurobiology will include scientific research and research-based teaching, and participation in international collaboration. In addition to giving lectures and courses, the professor will supervise undergraduate and postgraduate students and participate in the development of Bachelor’s, Master’s and doctoral programmes, in particular the Faculty’s Master’s Programme in Neuroscience (https://www.helsinki.fi/en/programmes/master/neuroscience) and the Doctoral Programme Brain and Mind (https://www.helsinki.fi/en/research/doctoral-education/doctoral-schools-…) which is hosted by the Faculty.

In addition, the appointee shall present documentation of international cooperation in the field of research that he or she represents.

An appointee to an assistant / associate professorship in the tenure track system (https://www.helsinki.fi/en/university/working-at-the-university/tenure-t…) shall hold a doctoral degree, have the ability to conduct independent scholarly work and have the teaching skills necessary for the position. In addition, applicants for assistant / associate professorships shall demonstrate their capability and motivation for an academic career through publications and other means.

When considering the applicant’s qualifications, attention will be given to scientific publications, teaching experience and pedagogical training as well as the applicant’s participation in doctoral education. Furthermore, account shall also be taken of the applicant’s activity in the scientific community, success in obtaining external research funding, international research experience and positions in Finnish and international organizations.

There is no requirement to be proficient in Finnish for foreign citizens, non-native Finnish citizens or citizens who have not been educated in Finnish or Swedish. To successfully attend to the duties of the position, the appointee must have good skills in English.
The professor’s salary will be based on levels 8−10 and the assistant / associate professor’s on level 7 of the job requirement scheme for teaching and research personnel in the salary system of Finnish universities. In addition, the appointee will be paid a salary component based on personal work performance. The gross monthly salary will be approximately 4500–8000 €, depending on the appointee’s career stage and scholarly qualifications.
