Doctoral Researcher position in immuno-oncology – University of Turku

We are looking for a highly motivated doctoral researcher to join the 3-year period ImmuDocs Doctoral Education Pilot project “Macrophages and neutrophils in cancer progression”, led by Professor Jyrki Heino, University of Turku.

Project description:

Malignant tumours consist of transformed cancer cells and of numerous other cell types including cancer associated fibroblasts (CAFs), tumour associated macrophages (TAMs), neutrophils, lymphocytes and endothelial cells. All cell types regulate each others’ phenotype and behaviour and cancer progression. Extracellular matrix (ECM) – mainly produced by CAFs – is an important component of the tumour microenvironment. ECM has a major impact on cancer cells, on the efficacy of immunotherapies and finally on the disease outcome.

We have developed new 3D spheroid-based methods to study the interactions of different cell types: human cutaneous squamous cellular carcinoma (cSCC) cells, CAFs and TAMs/neutrophils. Using e.g. mass spectrometry, RNA-sequencing, microscopy and cellular assays for proliferation, phenotype/plasticity and invasion the project aims to unveil the TAM and neutrophil based regulatory mechanisms of invasion and ECM remodelling. Furthermore, our preliminary results have revealed the remarkable plasticity of TAMs. The project aims to find molecular mechanisms used by CAFs and cSCC cells to regulate TAM phenotype.

In our recent paper we showed that the spheroid model can be used to successfully screen potential drug molecules (Siljamäki et al., Oncogene, 2023. PMID: 37864034). In this doctoral project the spheroids containing three different cell types will be used to test the effects of the inhibitors of critical signalling pathways. The model also allows testing of putative drug molecules targeted to macrophages and neutrophils.

Finally, we have recently established a method to combine laser capture microscopy to mass spectrometry and shown that it is possible to get detailed information about e.g. ECM composition in different tumour areas of paraffin embedded SCC tumours and their metastases. This in vivo information is critical for interpretation of the results from in vitro experiments focused on the effects of macrophages and neutrophils on the composition and organization of the ECM.

Orion’s Immuno-Oncology (IO) research team is looking to recruit an enthusiastic and motivated Immunology and flow-cytometry scientist to join the IO team. In this role you will contribute to IO target identification and validation, coupled with being responsible for supporting multi colour flow cytometry needs. In this role you have a unique opportunity to contribute in a range of treatment modalities ranging from small molecules, large molecules (bispecific & monospecific antibodies) and CAR T cells.

The responsibilities of this role include the following (including, but not limited to);

  • Design and perform in vitro immune assays and treatment with oncology drugs in conjunction with internal project teams.
  • Desing multi-colour flow cytometry antibody panels to detection and FACS sorting of a range of Immune cells.
  • Establish and validate human and mouse in vitro primary immune cells assays to define the mechanism of action of IO drugs.
  • Co-ordinate with the line-manager to ensure timely delivery of data and report if any issues encountered or foreseen.
  • Be responsible person for flow cytometry training for internal staff and regular maintenance of flow cytometry and cell sorter instruments.
  • Provide clear reports and presentations as required and participate in project team meetings and organisation meetings.
  • Working in close collaboration with other scientists and technicians.

This is a fixed-term contract of employment (parental leave) and location is Turku, Finland.

Requirements

To be a successful ideally you should have the following educational and professional work experiences:

  • PhD degree in Immunology or related field with 2+ years of relevant experience.
  • Strong and deep knowledge building multi colour panels, flow cytometry and cell sorter instruments
  • Strong in vitro research experience using primary immune cells assays is a must
  • Demonstrate ability to independently design, conduct and interpret the data from complex in vitro immune cell assays and flow cytometry is a must.
  • Demonstrate proficiency in isolation of immune cells and processing samples for multi-colour flow cytometry, multiplex ELISA, IHC, single cell RNA sequencing and gene expression analysis.
  • Competency in using graph pad prism, flowjo, nSolver software and other standard office software.
  • Excellent attention to details with strong communication skills.
  • Flexibility and willingness to learn new techniques and take additional responsibilities as a member of the research team.

The Centre of Excellence in Body-on-Chip Research (CoEBoC) is a multidisciplinary research consortium combining cell biology and engineering and aims at developing body-on-chips to model human tissue function.  The center has multidisciplinary expertise on human stem cells, biomaterials, sensors, microfluidics, biomodeling and bioimaging. CoEBoC produces new comprehensive knowledge on understanding tissue interactions, constructing complex in vitro tissue co-cultures and controlling their functionalities. We are looking for 6-16 Research Assistants to work on research and development projects within multidisciplinary research consortium. The open positions are related to ongoing research projects of CoEBoC concentrating on stem cells, cell differentiation, molecular biology, 3D culturing, biomaterials, sensors technology, microfluidics, microfluidic chips, biomodeling or bioimaging.

Monikudosmallintamisen huippuyksikkö on kuuden Tampereen yliopistossa toimivan tutkimusryhmän muodostama poikkitieteellinen tutkimuskeskus. RESEARCH TECHNICIAN/LABORATORY COORDINATOR (Genomics and molecular biology)

for the period of 1.2.2024 – 31.12.2025 (the starting date can be negotiated) to assist an international team in conducting molecular biology and genomic analyses e.g. DNA and RNA extraction, next-generation sequencing (e.g. bulk and single-cell RNA-seq), targeted SNP assay development and analysis, cell biology and histology techniques. Additional tasks may include ordering reagents, database maintenance, helping maintain experimental Atlantic salmon and rainbow trout populations, measuring and sampling fish and assisting other lab members as needed.

The successful candidate will likely have experience with at least some of the above-mentioned techniques and be enthusiastic about learning others. They will mainly work under the supervision of post docs and/or PhD students .We are looking for a highly motivated candidate with excellent organizational skills and good English communication skills. We appreciate a cooperative attitude and enthusiasm for learning new techniques and skills.

The salary is based on levels 6-8 of the job requirement scheme for other personnel in the salary system of Finnish universities. In addition, the appointee will be paid a salary component based on personal work performance. The position is offered with a six-month trial period.

The University of Helsinki Organismal and Evolutionary Biology Research Programme and the research groups of Prof. Jaakko Kangasjärvi , Dr. Maija Sierla and Dr. Cezary Waszczak are seeking to hire two highly motivated Postdoctoral Researchers to study Arabidopsis thaliana guard cell signalling.

Project Descriptions

The projects aim at molecular-level understanding of guard cell signalling and stomatal function in response to environmental stimuli. We also address connections between stomatal and vascular function. We use forward- and reverse-genetics, interactomics, and phosphoproteomics approaches coupled with plant physiology, molecular and cell biology and biochemistry to identify new components in guard cell signalling networks and further characterize the components we have previously identified.

Project 1: We have recently conducted a forward genetic screen aiming at saturation of Arabidopsis thaliana guard cell signalling network. Through an ozone-sensitivity approach combined with whole-rosette gas exchange analysis we identified over 50 novel mutants impaired in stomatal closure ( Sierla et al., 2018 ; Takahashi et al., 2022 ; Waszczak et al., 2023 ). One of the successful candidates will be involved in mapping and functional characterization of the newly identified mutants. This project will be performed in close collaboration between research groups of Prof. Kangasjärvi, Dr Maija Sierla and Dr Cezary Waszczak.

Project 2: We are using interactomics (Co-IP and TurboID) and phosphoproteomics approaches, and suppressor mutant screens with known stomatal regulators to further characterize the guard cell signalling pathways regulating stomatal movements in response to both abiotic and biotic stimuli. We also investigate the coordination of stomatal and vascular function. Our main focus is on the receptor-like pseudokinase GHR1 ( Sierla et al., 2018 ; Hõrak et al., 2016 ) and its novel interactors. One of the successful candidates will lead some of these research lines based on their expertise and interest in the group of Dr Maija Sierla.

The candidates will be integrated into an international research team of experts in plant stress biology and environmental signalling and participate in extensive collaborations, including mobility exchanges within Europe.

Qualifications:

  • doctoral degree in molecular or cell biology, plant biology, biochemistry or other relevant field
  • experience in plant physiology, molecular plant sciences and/or biochemistry (proteomics, interactomics, phosphoproteomics, kinase assays)
  • ability to conceive, execute, and complete research projects

  • excellent written and verbal communication skills

Työpaikkakuvaus

Dosentti Pieta Mattilan tutkimusryhmään haetaan tutkijatohtoria/erikoistutkijaa projektiin, jossa selvitetään virusantigeenien prosessointia ja esittelyä B-soluissa, tavoitteena ymmärtää paremmin tämän prosessin roolia ja siihen vaikuttavia tekijöitä rokotevasteessa. Valitun hakijan toivotaan aloittavan 1.3.2024 (tai sopimuksen mukaan). Tehtävä täytetään kaksivuotisena, mutta on mahdollista, että tehtävä jatkuu.

Ryhmämme tutkii B lymfosyyttien aktivaatiota, mikä mahdollistaa suojaavien korkean affiniteetin omaavien vasta-aineiden muodostumisen. Käytämme paljon moderneja mikroskopian ja proteomiikan työkaluja yhdistettynä soluaktivaatiokokeisiin sekä hiirimalleihin, joiden avulla pyrimme paremmin ymmärtämään B-solujen aktivaation säätelyä. Läpimenevänä tavoitteenamme on löytää uusia kohteita lääkekehittelyyn B-solulähtöisiin lymfoomiin tai immuunijärjestelmän häiriöihin, kuten immuunipuutoksiin tai autoimmuunisairauksiin, ja toisaalta löytää työkaluja tehokkaampien rokotteiden kehittelyyn.

Palkattava henkilö tulee työskentelemään osana EU-rahoitettua (Horizon Europe) projektia Yellow4FLAVI ( Deconstructing the protective immunity of yellow fever virus 17D to inform flavivirus vaccine design ). Yellow4FLAVI konsortio käsittää 13 partneria 7 eri maasta. Konsortiota koordinoi Prof. Giovanna Barba-Spaeth Institute Pasteurista, (Pariisi, Ranska). Meidän osuutemme on tutkia keltakuumeviruksen antigeenin prosessointia ja esittelyä B-soluissa. Työ sisältää erilaisten mikroskopiatekniikoiden käyttöä ja kehittelyä sekä soluaktivaatiokokeiden pystyttämistä kyseistä tarkoitusta varten tiiviissä yhteistyössä Ranskassa ja Saksassa sijaitsevien kollaboraattoreidemme kanssa.
Yellow4FLAVI projektin taustaa hieman: Aiemmin suurimmaksi osaksi tropiikkiin liitetyt flavivirukset, kuten Zika, dengue, Länsi-Niilinvirus ja keltakuume ovat laajenemassa globaaliksi terveysuhaksi. Ilmastonmuutos ja kaupungistuminen ovat edesauttaneet viruksia levittävien hyttysten alueiden laajenemista ja Euroopassa onkin viime vuosina havaittu useita paikallisesti leviäviä epidemioita. Keltakuumevirus 17D (YF17D) on heikennetty viruskanta, jolla saavutetaan rokotevaste, joka on ylitse muiden tehokkuudessa ja turvallisuudessa. Rokotteen menestyksestä huolimatta emme ymmärrä, mikä tekee siitä niin erinomaisen. Yellow4FLAVI konsortio pyrkii ymmärtämään näitä mekanismeja, yhdistäen viruksen rakenteen saavutettuun immuunivasteeseen, tavoitteena oppia kehittämään parempia rokotteita myös muita flaviviruksia vastaan.
