The Cell Biologist Metabolism supports drug discovery
projects with scientific input in the in vitro assay strategy and delivers key in vitro and ex vivo
decision-making data.
Major responsibilities:
• Independently apply existing in vitro and ex vivo assays for projects during different phase of
projects.
• Independently design and execute in vitro or ex vivo experiments
• Statistical analysis and interpretation of generated data
• Presentation of data at the team and project team level
• Document the work according to AstraZeneca standards, and prepare the preclinical
pharmacology part of regulatory documents as appropriate.
• Work collaboratively with in vivo, DMPK, etc scientists as required to ensure flow-scheme
connectivity and translatability between in vivo and in vitro models
• Work collaboratively within Bioscience and iMed to deliver on project key questions.
• Contribute proactively to a vibrant and innovative working environment where improvements
and new ideas are adopted and freely shared.
• Ensure compliance with corporate code of conduct guidance and other safety standards.
Minimum requirements
• Broad expertise in cell biology & biochemistry
• Experienced in establishing/performing cell-based assays.
• Cell culture, ELISA, western blot, and qPCR experience is a must

Extracellular
vesicles represent an exciting opportunity for delivery of complex
RNA-based drugs, and since 2015 we have been working with a team of
postdocs on developing new technologies for drug loading, targeting and
functional delivery using extracellular vesicles. We now have openings
for three new postdocs working on 1) delivery of mRNA, 2) targeting to
heart, and 3) developing innovative in vitro and in vivo
models of heart disease to explore the action and underlying biological
mechanisms of engineered extracellular vesicles in a therapeutic
setting. The internal postdoc work at AstraZeneca is supported through
collaborations with leading academic experts.
Myocardial
infarction (MI) results in severe damage to the heart muscle tissue with
scar formation and irreversible loss of cardiac function leading to the
development of heart failure. Encouraging results have been achieved
with cell therapy to regenerate the heart tissue and improve function,
but there is accumulating evidence that these beneficial effects are
mainly mediated by extracellular vesicles. We propose to study the
action of engineered extracellular vesicles from human IPS-CPCs (cardiac
progenitor cells) and from adult stem cells in vitro (angiogenesis, cardiomyocyte proliferation, apoptosis) and in rodent models of MI in vivo. We will engineer extracellular vesicles
to carry additional (known and novel) stimulatory factors to augment
their biological activity (e.g. VEGF-A, mRNA coding for genes driving
proliferation, etc.). We have developed novel cell-based and in vitro
technologies for EV loading with proteins and RNA, which will be
exploited in this project (focus for postdoc-1). We will explore targeting of extracellular vesicles to heart tissue (focus for postdoc-2) with known and novel ligands and examine improved drug exposure to heart tissue. We will multiplex in vivo
experiments with extracellular vesicles by bar-coding technology for
bio-distribution studies. In addition, we have a separate recruitment
ongoing in CVRM for a 3rd AZ postdoc working on the development of novel
in vitro and in vivo regenerative models of heart
disease. The three postdocs will work closely together on the shared
goal of developing novel opportunities around the use of engineered
extracellular vesicles for therapeutic treatment of heart disease. As a
postdoc you will also get the opportunity to try novel techniques and
ideas. We will also support development of leadership skills and deeper
understanding of drug development process.
Education and Experience required:
Essential:
- A PhD in biological sciences with a strong cell biology or biochemical focus.
- A strong scientific track record, as evidenced by published papers in high quality peer-reviewed journals.
- Excellent
organizational, communication and interpersonal skills, combined with
the ability to work collaboratively across boundaries and problem solve

Subject field and description of the position

The doctoral student project and the duties of the doctoral student
The doctoral education project will be carried out within the Horizon2020 project FREIA (http://www.freiaproject.eu/
), which rises from a concern that environmental factors can affect a
woman’s chances of becoming pregnant. Hence, the ambition of FREIA is to
develop novel identification and screening approaches for endocrine
disrupting chemicals (EDCs) that target female
fertility. The aim of this doctoral education project is to study the
effects of EDCs on human ovarian follicle growth in vitro and identify
associated adverse outcome pathways. Cross-species validation will be
carried out by comparing the findings to rat studies
carried out in FREIA. In addition, ovarian transcriptomics datasets will
be used to understand possible age-dependent differences in chemical
toxicity. The work will require visits to laboratories of co-supervisors
in Finland, Denmark and Estonia.
The
duties of the doctoral student include cell and tissue culture, gene
expression analyses and histological analyses. In addition, the student
will assist in collection
and analyses of rat ovaries and oocytes. The project also involves
analyses of RNA-seq datasets. Techniques utilized will include in vitro
culture of human ovarian tissue samples, isolation and culture of
primary ovarian cells, immunostaining, fluorescent microscopy
and histomorphometric analyses, as well as bioinformatics analyses in R.

will join a collaborative immuno-oncology project funded by an Academy
of Finland project grant of Dr Jeroen Pouwels that seeks new ways to
sensitize breast cancer to immune checkpoint
inhibitors. The very translational research group is part of the
Translational Cancer Medicine Research Program of the Medical Faculty’s
Research Programs Unit (RPU) and is aligned with the Finnish Flagship
Programme iCAN. A novel method to grow 3D cultures
of intact fragments of patient-derived tissue (Patient-Derived Explant
Cultures), which preserves the human tumor microenvironment including
the immune-contexture, reflects inter- and intra-patient tumor
heterogeneity and, importantly, can assess the efficacy
of novel treatment regimens on real human tumors (Haikala et al., Nature

Scientist, Neuroimmunology and immunohistochemistry
We are looking for a scientist with proven track of experience in ex vivo molecular and cellular neuroscience.
The
position requires expertise in immunology and immunohistochemistry in
basic science and preferably drug discovery using preclinical models of
CNS.
In
this position you will be independently leading projects focusing to
relevant ex vivo endpoints of efficacy in animal models, including e.g.
PD, AD, MS and injury-triggered animal models.
You
have a strong expertise in various types of ex vivo analysis and
interpretation of biomarkers derived from preclinical in vivo models.
You have strong background e.g. in multiparameter flow cytometry,
immunohistochemistry, cytokine profiling, supporting immune function
assessment.
You
are independently able to manage planning, coordination, supervision
and execution of the ex vivo part of client projects and ensuring the
scientific excellence including data analysis and reporting of the
results. Scientist will be working closely with other teams and clients.
In addition to client projects, scientist will be involved R&D
activities including validations of new markers and assays to support ex
vivo readouts.
Qualifications and Experience:
- Higher education degree in biological science (MSc) with 3+ years of experience, preferably PhD degree
- Pharmaceutical industry, Preclinical Contract Research Organization or academic post-doctoral experience will be an asset.
- Good problem solving skills, ability to coordinate and prioritize
- Scientific expertise with demonstrable understanding of ex vivo techniques and neuroimmunology as a component in CNS diseases
- Understanding of key features of in vivo pharmacology studies and drug development process in neurology and psychiatry
- Hands on experience in development, optimization and validation of immunohistochemistry targets in CNS tissues
- Hands
on experience in development, optimization and validation of
multi-color flow cytometry, data analysis and troubleshooting using
multiparametric data analysis
- Experience
in other immunological techniques like in vitro cell cultures, Luminex,
ELISA, gene expression analysis (qPCR, QuantiGene Plex) will be an
asset
- Fluent oral and written English, excellent communication skills, interpersonal and teamwork abilities
- Ability to mentor and train staff will be an asset

PhD project in inflammation and infections group
PhD
project is available in the research group led by Dr. Karita Haapasalo
at the Department of Bacteriology and Immunology, Haartman Institute. We
are looking for an ambitious and enthusiastic
MSc of MD with a research interest in immunology and inflammatory
diseases. The project is suitable for a applicant with a knowledge of
immunology, microbiology, biochemistry, cell biology or other relevant
field. After a four months trial period there is an
opportunity to continue the project and apply for PhD studentship.

lab studies mitochondrial morphogenesis and dynamics. More
specifically, we focus on the molecular mechanism of mitochondrial
membrane architectural organization and factors involved in regulation
of mitochondrial shape, dynamics, and their functional link to
mitochondrial diseases.
We
